United missed out of the continental championship last season after a disappointing spell under former manager David Moyes. The coach Louis van Gaal revamped Red Devils will be looking to see off the 13-time Belgian champions in the tie scheduled to be played on Tuesday 18 and Wednesday 26 August.
It is the first time the 20-time English champions are playing in the preliminary rounds of the flagship co competition since 2005, when they mauled Hungarian side Debrecen 6-0 on aggregate scores.
Brugge sealed a 4-2 (aggregate) come-back win over Greek club Panathinaikos in the third qualifying round to book a slot in the play-offs. It will be the first competitive match between the Premier League club and the Belgians, who were European Cup runners-up in 1978, losing to Liverpool in the final.
